I'm trying to edit the coordinates using plane maker so the winch line is aligned with the winch object while it's in the extended operating position ('winde' on custom slider 6). Changing the Lat, Long and Vert coordinate values in the weight and balance menu of plane maker does not seem to have any effect. Are these values independently set within your EC135 Plugin? If so is there any way to adjust them so the winch line is correctly positioned and aligned with the winch?

You are right, the coordinates are controlled by the plugin. If you have a cargo hook it is another position for the sling load then with a winch.
Unfortunately the winch is not yet ready for use. One fine day the position will change with the angle of the winch, that will be controllable like in reality. Please be patient.

The plugin already automatically changes the coordinates of the rope suspension according to the extension angle of the winch boom.
You can easily test this using dataref editor/tool and looking at the values of the following datarefs while issuing commands ec135/hoist/out or ec135/hoist/in:
sim/aircraft/overflow/jett_X
sim/aircraft/overflow/jett_Y
sim/aircraft/overflow/jett_Z
